This research focuses on the study of the effect of electroporation,
a technique that reversibly permeabilizes membranes, on human cadaver
skin in enhancing the delivery tacrine, a reversible cholinesterase
inhibitor used for improved cognition in Alzheimer’s patients.
Tacrine is a potent drug that is only available in oral dosage form.
The drug exhibits high first pass metabolism, necessitating frequent
and high dosing. Transdermal delivery of the drug can reduce the effects
of first pass metabolism and can eliminate the need for frequent and
high dosing of the drug, thus improving patient outcomes. |

This project will focus on the pharmacokinetic and pharmacogenetic
differences between Caucasian and African-Americans, in particular
Somali African-Americans. Differences in pharmacokinetics have been
identified among racial groups. The standard practice in medicine
is a “one dose fits all” approach, which may not be appropriate
for all patients. This project will identify the particular mutations
present in CYP2C9, 2C19, or 3A4 in individuals and determine if there
are differences in the pharmacokinetics of the older antiepileptic
drugs according to isoenzyme mutation. |
