Focus of Research: This project examines regulation of an enzyme involved in production of lysophosphatidic acid (LPA). LPA increases growth of ovarian cancer cells, and is elevated in ovarian cancer patients. The goal of the project is to determine whether autotaxin activity is stimulated by epidermal growth factor (EGF) in human ovarian cancer cells. If not, other growth factor regulated phospholipases must be involved in growth factor-mediated LPA production.

Focus of Research: To determine the influence of the inhibition of 3,4-methylenedioxymethamphetamine (MDMA, Ecstasy) metabolism via inhibition of cytochrome P450 on the formation of the metabolite 3,4-methylenedioxyamphetamine (MDA). The research will evaluate the influence of MDMA as the causative agent in generating the hyperthermic response seen after administration of MDMA. The inhibition of the formation of MDA will allow the hyperthermic response to be evaluated in the presence of MDMA alone.

It is unknown whether this affects the propensity to exhibit drug-drug interactions. This project will examine whether the *10 and *17 alleles are more or less susceptible to drug interactions than the wild type enzyme using a variety of substrates and inhibitors.

Focus of Research: To custom- develop a nucleic acid aptamer drug carrier system using Systemic Evolution of Ligands by Exponential enrichment, a.k.a. SELEX. This carrier will allow drug targeting and controlled release of the molecule being delivered. Allosteric regulation of drug release will closely mimic the control mechanisms of naturally occurring physiological feedback loops, creating stable serum levels and decreasing the chance of adverse events.

Focus of Research: To evaluate the effects of aromatase inhibitors on: brain aromatase activity, cognitive performance, and basal forebrain cholinergic function. Use of aromatase inhibitors is becoming increasingly prevalent in the treatment of breast cancer and aromatase is also expressed in the brain. Studies suggest that local conversion of testosterone to estradiol in the brain can play an important role in neuroprotection, synaptic plasticity, and cognitive function (due, at least in part, to effects on basal forebrain cholinergic neurons).

Focus of Research: This research focuses on atrial fibrillation (AF), including the efficacy and safety of amiodarone for prevention of AF following non-cardiac thoracic surgery and the influence of prevention of AF following non-cardiac thoracic surgery on post-surgical duration of stay in the ICU. Pre-surgical plasma concentrations of B-type natriuretic peptide (BNP) will be monitored to determine whether BNP predicts development of AF after non-cardiac thoracic surgery. BNP, epinephrine, and/or norepinephrine post-surgical concentrations will also be monitored for elevation in patients that develop AF after non-cardiac thoracic surgery.

Focus of Research: To encapsulate small inference RNA (siRNA) in thiolated gelatin nano-particles for rapid intracellular delivery in response to glutathione. The results of this study will be used to optimize formulation design for maximum gene silencing in vitro.
